This was a great experience, definitely worth the full day and the price. Our El Yunque guides, Daniel and Rene, were fantastic. The jungle hike involves wading waist deep in the river so I'd recommend very supportive water shoes or sneakers you don't mind ruining. We went with water shoes with ok support (could have been better) and it was doable. The hike is moderate difficulty, but should be something anyone who is generally active can handle.
The bio bay portion of the tour was handled by a different tour company, Kayaking Puerto Rico. My wife and I really enjoyed the kayaking and we were able to see the bioluminescence with the tarp. Unfortunately it was weaker when we went, but we were still happy with the experience. Kayaking back from the bay through the mangroves in the dark was quite challenging. We got the impression that some in our group had a hard time and didn't enjoy that part. There was basically no light besides tiny red LEDs on the front/back of each kayak that didn't light up their surroundings at all. Our guides would occasionally use a flashlight if someone got stuck, but otherwise we were in pitch dark. My wife and I still enjoyed this part of the experience as we have a decent amount of experience kayaking, but it might be worthwhile to consider additional lighting for those with less experience.
